The treatment of erectile dysfunction (ED) is complex and not completely understood, and its causes remain unknown. The treatment of ED, such as Viagra, is limited by its side effects and tolerability, which may include the common side effect of sildenafil. In this study, we investigated the safety of sildenafil citrate, a phosphodiesterase-5 (PDE5) inhibitor, in patients with ED using an ED patient data bank.
Methods:This double-blind, placebo-controlled, randomized, single-center study included 17 patients with ED according to the Global Initiative for Sexual Medicine guidelines. Patients were randomized into treatment groups (n = 17) and placebo (n = 17). The study protocol was approved by the University of Texas Health Science Center (UHCSPC) institutional review board and the study participants signed the informed consent.
Results:There was no statistically significant difference in the adverse event rate in the placebo group in comparison with the sildenafil group (45.3% vs. 43.4%, respectively), the difference in the time to first sexual episode, the time to first sexual episode, the duration of erectile dysfunction (ED) (4.7 vs. 3.1 minutes, respectively), or the rate of sexual intercourse (11.1% vs. 9.4%, respectively). The difference in the time to first ED episode (3.4 minutes vs. 6.3 minutes, respectively) was statistically significant. The incidence of mild and moderate erectile dysfunction was similar in the sildenafil and placebo groups (0.6 vs. 0.9%) (p = 0.4 and 0.1, respectively). The number of patients who received at least one dose of sildenafil was greater in the sildenafil group than in the placebo group (1.7% vs. 0.8%).
Conclusion:In the present study, the safety profile of sildenafil was similar to that of sildenafil citrate. It should be noted that this is an open-label, single-center, placebo-controlled study, so it should be conducted under strict supervision from healthcare professionals.
